This can happen after a Blackmagic driver upgrade, especially when they are 2x DeckLink cards from different models in the computer. For example, if the PC has 1x DeckLink Studio 4K and 1x DeckLink 8K Pro, the second card won't be detected in Desktop Video Setup.

Usually, Blackmagic detects the second card and suggests an update automatically.
If not, open the Command Prompt by typing CMD on Windows.
Change repertory by typing
cd C:\Program Files\Blackmagic Design\Blackmagic Desktop Video
Then type the command (note : it's an L like Lima)
DesktopVideoUpdateTool.exe -l
Find the missing DeckLink card and copy its ID. In the example above, the missing card is a Decklink Quad 8K Pro and its ID is G808e2f50.
Type the command
DesktopVideoUpdateTool.exe -u -d <id of the missing decklink card>In the example above, the command will be
DesktopVideoUpdateTool.exe -u -d G808e2f50
Let the update run. This can take a few minutes to finish (around 5 to 10min). When it finishes, restart the computer.
Open Desktop Video Setup and check your cards again. You should see the missing card now.
If it still does not work, you may have to disable the Resizable Base Address Register option in the BIOS.
BIOS > Advanced > PCI Subsystem Settings > Re-Size BAR Support > Disabled
